Effect of mAbs on adhesion of ICAM-1 transfected L cells to purified CD11b/CD18

ICAM-1-expressing L cells were detached, washed, and resuspended (5 × 106/ml) in PBS containing 2 mM MgCl2 and 2% heat-treated BSA as described in Materials and Methods. Cells were preincubated for 30 min with no antibody (No mAb) or with one of the anti-ICAM-1 mAb above (20 µg/ml, 4°C). Subsequently, cells were washed, resuspended (106/ml), and allowed to bind to 6-cm petri dishes coated with purified CD11b/CD18 for 50 min (37°C). Unbound cells were washed free and adherent cells were quantitated as described previously (22). Bars = mean ± SD of adherence normalized with respect to binding in the absence of antibody (100%); one of three experiments.
