Kinetic analysis of the glial cell-specific gene expression in the forebrains and cerebella of Ngsk Prnp0/0 mice. (a) The levels of transcripts for GFAP, LM, and IP3R1 were compared between Ngsk Prnp0/0 and Prnp+/+ brain tissues at different ages (wk). 10 µg of total RNA extracted from the pooled tissues were subjected to Northern blotting. The integrity and quantity of the RNA were verified with GAPDH probe on the same membrane. (b) Kinetics of the mRNAs for GFAP, LM, and IP3R1 were compared between Ngsk Prnp0/0 (closed circles) and Prnp+/+ (open circles) mice. The intensities of Northern blot signals shown in (a) were quantified using BAS 5000, standardized with GAPDH mRNA level, and expressed as arbitrary units in comparison with the value of Prnp+/+ mice at 3 weeks of age.
