Correlation of insulin-stimulated mTORC1 activity or glucose transport to whole body insulin sensitivity (QUICKI). Adipocytes from 26 nondiabetic subjects (Supplemental Table S1) were examined. (A) Glucose transport into isolated adipocytes was determined, in triplicate, as uptake of 2-deoxy-d-glucose with (filled circles) or without (open circles) preincubation with 10 nmol/L insulin for 15 min. (B–D) The effect of insulin on the phosphorylation of S6K1 (B), S6 (C), and IRS1 at serine-307 (D) were determined in isolated adipocytes after incubation with or without 10 nmol/L insulin for 10 min, when cells were subjected to SDS-PAGE and immunoblotting with antibodies against phosphothreonine(389)S6K1 (B), phospho-serine(235/236)S6 (C), or phospho-serine(307, human sequence)IRS1 (D). Results are given as fold over of controls without insulin, mean of triplicates.
